摘要:目的:研究南澳岛海域亨氏马尾藻多糖的提取工艺,并对其进行纯化和鉴定。方法:用热水提取多糖,乙醇沉淀,并用正交实验确定提取的最佳条件,Sevage法去蛋白,用CTAB结合不同浓度盐离子溶液对多糖进行分级。SephadexG200柱层析及聚丙烯酰胺凝胶电泳进行纯度鉴定。硅胶薄层层析分析多糖中单糖组分。结果:最佳提取条件:100℃,pH7,提取4h,得率可达22.7%,分级沉淀后得四组分F1、F2、F3、P,纯度鉴定证明其中F1、F2、P为均一组分,薄层层析发现其中含有岩藻糖。结论:亨氏马尾藻多糖的分离、纯化和鉴定能够为该多糖的进一步开发利用提供一定的理论依据。

Isolation, Purification and Identification of Polysaccharides from Sargassum Henslowianum Ag

Abstract:Purpose: To study the isolation, purification and identification of polysaccharides from Sargassum henslowianum Ag of Nanao Island. Methods: The polysaccharides were isolated with hot water, and precipitated with ethanol. We could get the best condition with perpendicular experiments. The protein in polysaccharides was removed by sevage way. The polysaccharides were graded by CTAB and various salt solution. Their purities were identified by Sephadex G 200 and polyacrylamide gel electrophoresis. Their components were analyzied with thin-layer chromatography. Results: The best isolating condition was 100℃, pH7 and isolating 4h. The polysaccharides were graded into F1, F2, F3, P and it was approved that F1, F2, P were homogeneous and there was fucose in them. Conclusion: The isolation, purification and analysis of polysaccharides from Sargassum henslowianum Ag could provide a theoretical foundation for further development and utilization.
