黄岑中酚性甙类对小鼠放射损伤防护作用的研究

摘要目的:探索黄苓中的放射损伤防护作用有效成分。方法:黄芩水提液酸化保温(80℃)50分钟,滤去析出的黄芩黄酮类(FSBG),水溶液用经典分离法得一沉淀物(Ⅰ)、化学分析其结构类型;用60Co7.7Gy~8.7Gy全身照射纯系NIH雌性小鼠,照前35~45min每鼠腹腔注射(Ⅰ)1g/kg或2g/kg,观察生存防护效力、30天存活率和照射4.0Gy(2g/kg)后第4天和第12天的WBC、PLT、RBC和HGB。结果:(1)FSBG和(Ⅰ)均对三氯化铁、Molisch、Fehling试剂呈阳性反应,前者对浓硫酸、碱试液和盐酸镁粉呈阳性而(Ⅰ)呈阴性;(2)每鼠1g/kg和2g/kg剂量均显著提高生存防护效力(1.35~2.08治疗组30天存活时间和对照组30天存活时间之比值)和30天存率(治疗组为8.33~41.46%,对照组为0~2.50%),2g/kg能明显增加4.0Gy照后第4天和第12天的WBC、PLT、RBC和HGB。结论:(1)对浓硫酸等的呈色反应差异表明(Ⅰ)和FSBG化学结构不同;对FeCl3等阳性反应提示(Ⅰ)的主要成分为酚性性甙类;(2)GSBG有显著的放射损伤护作用;(3)本研究为黄芩的新应用提供了重要依据。

Study on the protective effect on radiation injury of mice by glycoside with phenolic hydroxyl group from Scutellaria baicalensis Georgi

AbstractObjective:The effective principle with protective effect on radiation injury from Scutellaria baicalensis Georgi (SBG) was found and studied.Methods:The heated extracted aqueous liquid from SBG was acidified and kept hot for 50 minutes in 80℃ , the flavonoids of SBG (FSBG) were precipitated and removed by filter.A precipitate (Ⅰ) from the filtered solution was obtained by the classical separation process.Tests were conducted on mice to detect whether (Ⅰ) is effective for protection against γ -ray irradiation.Intraperitoneal injection(ip) of (Ⅰ) 1 g/kg and 2 g/kg in NIH female mice 35~45 minutes befores 7.7~8.7 Gy of whole body 60Co irradiation were conducted,the survival effects,30 days survival rate and in case of ip of (Ⅰ) 2 g /kg the WBC,PLT,RBC and HGB were observed on 4th and 12th day after 4.0 Gy exposure.Results:Positive reactions were shown with following chemical reagents:ferric chloride,Molisch,Fehling reagents on FSBG and (Ⅰ),however,concentrated sulfuric acid,alkaline reagents and hydrochloric acid-Mg powder showed positive reaction on FSBG,but negative on (Ⅰ).Intraperitoneal injection of (Ⅰ) 1 g/kg and 2 g/kg increased the survival effects(1.35~2.08,the ratio of mean survival time in a treated group to that of the control group in 30 d after irradiation) and 30 days survival rate (8.33~41.46% in treated and 0~2.50% in control group,respectively) significantly in comparison with control mice.In cases of ip of (Ⅰ) 2 g/kg the WBC,PLT,RBC and HGb were all increased on 4 th and 12 th day after 4.0 Gy exposure as compared with control.Conclusions:1 The different coloration to chemical reagents (concentrated sulfuric acid and etc.) showed that there are different chemical structures between(Ⅰ) and FSBG.The positive reactions (ferric chloride and etc.) suggested that the main constituent of (Ⅰ) was a chemical type of glycoside with phenolic hydroxyl group. 2 The glycoside with phenolic hydroxyl group exhibited good radioprotective effect against whole-body lethal irradiation of mice.3 The study may be considered as new application of SBG.

前文报道黄芩(ScutellariabaicalensisGeorgi,SBG)水提物有显著的放射损伤防护(RP)作用[1],其功效与作者研制并经动物实验及临床应用效果显著的RP-2[2~6](黄芩为主药)相一致。本文报道进一步分离出一种黄芩酚性甙类(GlycosideofSBC,GS-BG)及其RP作用的研究结果。

1材料和方法

1.1GSBG的分离和化学检测

黄芩(产地河北省,购自广州赵秀药材分公司)水提液用盐酸调pH1~2。在80℃下保温50min,室温静置,滤去芩黄酮类(FlavoinoidesofSBG,FSBG)沉淀。滤液用经典分离法获得一沉淀物简称(Ⅰ);将FSBG和(Ⅰ)进行化学检测以鉴别其化学结构类型。

1.2动物实验和观察指标

1.2.1动物为纯系NIH雌性小白鼠,广东省卫生厅医用实验动物场提供(广东省实验动物合格证97A017号和98A032号;广东省实验动物条件检测合格证97B008号98B013号),体重18~22g。

1.2.2(Ⅰ)对小鼠照射后30d存活的影响:动物随机分为(Ⅰ)+照射组,照射组和生理盐水正常组(正常饲养,不作任何处理),照前35~45min前两组分别腹腔注射(Ⅰ)和同等容量的生理盐水。第1至第3次/实验(Ⅰ)的剂量为1g/kg,第4至第6次实验为2g/kg。第1至第3次每次实验3组动物各12只,第4次各14只,第5次各13只,第6次(Ⅰ)组14只,照射组和正常组各13只,观察:①生存防护效力(Ⅰ)+照射组30d平均存活天数/照射组30d平均存活天数)[7];②30d存活率。

1.2.3(Ⅰ)对小鼠照射后造血系统的影响:小鼠42只,随机分为3组:正常组(条件同上)12只,(Ⅰ)+照射组和照射组各15只。(Ⅰ)的剂量为每鼠2g/kg,照射前给药。照射后第4天和第12天取血,在Cell-Dyn1600血细胞自动分析仪上测定,第12天取血后颈椎脱臼处死小鼠秤体重、脾重和胸腺重。

1.2.4醋酸扭体试验:小鼠24只,体重20~22g,随机分为对照组和(Ⅰ)组各12只,分别腹腔注射生理盐水和(Ⅰ)1.5g/kg,30min后,腹腔注射0.21mol/L醋酸0.01ml/g体重,记录20min内两组小鼠扭体次数。

1.2.5对戊巴比妥钠阈下催眠剂量的影响:小鼠24只,体重20~22g,随机分两组各12只,分别腹腔注射生理盐水和(Ⅰ)1.5g/kg,30min后两组均腹腔注射0.12mol/L戊巴比妥钠0.01ml/g,观察30min内两组翻转反射消失的鼠数和持续时间。

1.2.6照射条件:动物用Theratron780型60Co放射治疗机全射照射1次,剂量率为1.02~1.04Gy/min。照射量7.7~8.7Gy观察30d存活影响;4.0Gy观察造血系统的变化。源皮距为80cm,照射野为25cm×25cm。

1.3统计方法

用两组t值进行统计。生存防护效力的P值是计得30d平均存活天数的P值后,据文献[7](见文中1.2.2)的计算式求出。

2结果

2.1(Ⅰ)的化学检测结果

(Ⅰ)和FSBG对三氯化铁、α-萘酚(Molish)、碱性酒石酸铜(Fehling)试剂呈强阳性反应。其酸水解液有絮状沉淀物产生且对Fehling产生的砖红色沉淀比水解前显著增多,提示两者均为酚性甙类;FSBG对盐酸镁粉微红、反应后溶液黄色。对浓氨水(纸片上)呈深棕黄色、在2537Å和3650Å紫外光下显深棕色。对1%碳酸钠(纸片上)呈淡棕黄色、在同上紫外光下显深棕色。对氢氧化钠液呈橙棕色。对浓硫酸显深亮黄色。对醋酸镁(纸片上)呈淡黄色。而(Ⅰ)对以上试剂均呈阴性反应;又FSBG对醋酸铅产生橙黄色絮状沉淀。对碱式醋酸铅产生橙红色絮状沉淀、放置后上层液黄色混浊。而(Ⅰ)对前一试剂产生棕色絮状沉淀,对后者产生橙棕色絮状沉淀、放置后上液无色澄明;FSBG对各种试剂显示了其黄芩黄酮类的特征性反应,而(Ⅰ)则有显著差异,提示(Ⅰ)的化学结构与FSBG不同,推定其为黄芩中的一种酚性甙类(GSBG)。

2.2(Ⅰ)对小鼠照射后30d存活的影响

从表1可见,生理盐水正常组小鼠30天存活率为100%且体重明显增加(26~29g)。照射组呈现食欲下降、消瘦、活动减少、松毛、稀便等随之死亡的时间比GSBG组显著提前,GSBG组的30天存活时间、存活率均明显高于照射组,其生存防护效力高达2.08,存活率为照射组的16.58倍。

2.3对造血系统的影响

和照射组比较:GSBG组在照射后第4天,其红、白细胞(含嗜中性和中间细胞)、血色素和血小板均有统计学显著差异,在第12天,以上各项(淋巴细胞亦P<0.05)多为显示差异有显著或非常显著意义(见表2)。照射后第12天,胸腺指数与照射组(3.22±0.76)比较,(Ⅰ)+照射组为4.00±0.71,正常组为4.06±0.35,t分别为2.57和2.47,P值均<0.05。两组脾指数无统计学显著差异。
