血脂康对家兔血管成形术后内膜增殖及内皮功能的影响(摘要)

目的:本研究应用实验性家兔动脉粥样硬化后血管狭窄模型,经球囊血管成形术后,观察应用血脂康对血管内膜增生及内皮功能保护的作用。

方法:日本纯种雄性大耳白兔36只,体重2.5±0.5 kg,月龄6~8个月,予1.5 g/(kg·d)高胆固醇饮食喂养4周后,穿刺右股动脉并导入球囊导管至肾动脉下方,充盈球囊剥脱动脉内皮。继续高脂喂养4周,经左股动脉插管,行腹主动脉造影,对狭窄部位行血管成形术,再经造影证实扩张满意为止。共成功建立30只家兔动脉粥样硬化后血管成形术模型,随机分为对照组、高脂组和治疗组[血脂康0.8 g/(kg·d),血脂康为北大维信公司生产];继续喂养4周后气栓处死。实验前后检测血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、血浆内皮素-1(ET-1)和血清一氧化氮(NO)。动物处死后,截取成形术部位血管标本,行弹力纤维染色,利用计算机图像分析系统分别测出新生内膜面积(NEA)、中膜面积(MA)和管腔面积(SA),并计算NEA/MA的比率。

结果:所有3组TC、TG、LDL-C、ET-1高脂喂养4周后均明显升高(P<0.05),而NO呈下降趋势;12周后,TC、TG、LDL-C、ET-1在治疗组中下降,低于第8周水平(P<0.05),在高脂组中保持增高,与对照组比P<0.05;治疗组在12周后明显下降,低于第8周水平,与对照组相比明显下降(P<0.05),而NO水平则明显升高,与对照组比较P<0.05。病理形态学观察NEA、NEA/MA比值的改变为:血脂康治疗组<对照组,对照组<高脂组,组间比较P<0.05。

结论:血脂康具有抑制家兔血管成形术后内膜增殖和保护内皮功能的作用。

The effect of Xuezhikang on Neointimal Proliferation and Endothelial Function after angioplasty in Rabbits (Abstract)

objective: To investigate the effects of Xuezhikang (XZhK, a Chinese patent medicine used for blood-lipid lowering) on the neointimal proliferation and endothelial function after angioplasty of the modelled rabbits.

Methods: angioplasty was performed on 30 modelled rabbits of atherosclerotic stenosis in celiac artery. After being fed with cholesterol-supplemented diet for 8 weeks, they were then randomly divided into the control group, high-cholesterol group and XZhK group. After an additional high-cholesteroldiet for 4 weeks, the local veassels were collected for morphological observation, and blood lipid, nitric oxide (NO) and endothelin-1 (ET-1) were also measured.

Results: the morphological observation showed that the neointimal area in the XZhK group was less than those in control group (p<0.05), and that in control group was less than in high-cholesterol group (p<0.05). The serum total cholesterol (TC)、 triglyceride (TG) and ET-1 levels were increased and the serum NO level was decreased after high-cholesterol diet. however, as compared with the control group and high-cholesterol group, there was more remarkable decrease in TC、 TG levels and increase in NO level in XZhK group.

Conclusion: xZhK had the effects of inhibiting the neointimal proliferation and of protecting the endothelial function after angioplasty in rabbits.
