一、XAP筛选的方法
迄今为止,有关XAP筛选的方法多采用酵母双杂交体系(yeast two-hybrid system),其基本原理是将HBV X基因与酿酒酵母GAL4蛋白的结合结构域基因融合构成杂交体,将来自人体细胞的cDNA文库同转录激活结构域基因融合成一个杂交体cDNA文库,然后将含X基因的杂交体和杂交体文库一起转化酵母,通过检测酵母中报告基因是否表达及其表达水平,从而在cDNA文库中筛选出与pX相互作用的XAP基因。
目前酵母双杂交体系的实验方法已逐渐成熟,主要步骤包括构建并检测钓饵质粒,筛选作用融合蛋白的质粒库,排除假阳性及对真阳性分析测序等[3],其中假阳性的排除与真阳性的鉴定至为重要。由于酵母双杂交体系研究蛋白之间相互作用的整个过程只对核酸进行操作,不需分离纯化蛋白的过程,且蛋白之间的相互作用发生在酵母细胞这一自然的状态下,因此反应的灵敏度较高。
二、XAP的研究现状
运用酵母双杂交体系,迄今已筛选出多个能与pX相互作用的XAP,并对其序列与性质进行了初步分析。……
