肝细胞色素P450 2E1在实验性肝纤维化组织中的表达

摘要:目的研究肝细胞色素P4502E1在实验性肝纤维化组织中的表达。方法8只Wistar大鼠,建立四氯化碳(CCl4)肝纤维化模型,用免疫组织化学方法观察肝纤维化模型肝组织中肝细胞色素P4502E1。结果正常肝组织内,CYP2E1表达仅见于中央静脉周围区,主要表达于肝腺泡Ⅲ区,2~3层细胞厚,肝细胞浆和细胞膜可见CYP2E1表达。在CCl4模型肝组织中,CYP2E1表达的强度增加,分布的方式亦发生改变,可见于肝腺泡Ⅰ和Ⅱ区,与肝细胞脂肪变分布相一致,在假小叶的肝细胞中不见其表达。CYP2E1在肝细胞内分布以胞浆型为主,亦可见膜型和核型。8只实验动物肝纤维化组织中,2只强阳性,3只中等程度阳性,3只弱阳性。结论CYP2E1表达与肝纤维化的发生有关,动物个体间存在CYP2E1多态性。

Expression of hepatic cytochrome P450 2E1 in experimental liver fibrosis

Abstract:Objective To study expression of hepatic cytochrome P450 2E1 in experimental liver fibrosis.Methods Experimental liver fibrosis model of rats was induced by carbon tetrachloride subcutaneous injection,and expression of hepatic cytochrome P450 2E1 was evaluated with immunohistochemistry by using a monoclonal antibody.Results In the normal rats,CYP 2E1 was localized within a rim of hepatocytes two to three cells wide around the terminal hepatic venule in acinar zone 3.The intensity of CYP 2E1 expression increased in the model,the acinar distribution of CYP 2E1 followed closely the distribu-tion of steatosis,sometimes extending through acinar zone 3 to zones 2 and 1,the hepatocytes in the pseudolobule weren’t found.The intensity and distribution of CYP 2E1 expression between rats of model differed.In 8 rats,2 rats were + 3,3 were + 2,3 were + 1.Conclusion CYP 2E1 expression was associated with pathogenesis of liver fibrosis.There were genetic poly-morphisms of cytochrome P450 2E1 between rats.
