DNA content study of normal hepatocytes,large cell change hepatocytes,hepatocellular carcinoma and hepatocytic peri-cancerous change in pericancerous parenchyma with image cytometry

Abstract:Objective To investigate the DNA content and morphologic characteristics of normal hepatocytes(NH),large cell change hepatocytes (LCC),hepatocellular carcinoma (HCC) and the hepatocytic pericancerous change (PC) in peri-cancerous parenchyma.Methods By use of histochemistry staining of DNA and image cytometry.Results In 10 cases of PC group,the DNA index (DI,1.99±0.34) were higher than that in 4 cases with NH group (0.98±0.12 ) and 8 cases with LCC group (1.46±0.19),P<0.01,the DNA ploidy pattern was also different as compared with that in NH and LCC group(P<0.05).The nucleus surface area and nuclear circumference (74.01±52.3, 63.89±33.9) were lower than those in 26 cases of HCC group (95.62±95.21,76.33±62.91),P<0.01.Among the PC group,80% (8/10) were of aneuploidy pat-tern,more than that seen in HCC group 53% (14/26),20% (2/10) were of polyploidy pattern,less than that in HCC group 26.92% (7/26),P<0.05.In the PC group,the hepatocytes were rich in nuclear chromatin and irregular nuclei shape,the cy-toplasmic cosinophilia or basophilic showed transitional change of HCC,replaceing growth pattern showed similar with HCC.Conclusion The abnormal DNA content,DI and the ploidy pattern of PC hepatocytes which is diffuse mass-like distribution and adenomatous arrangement showed precancrosis celles.
