白细胞介素13基因在人肾组织的表达及其意义

摘要:目的探讨白细胞介素13(IL-13)基因在原发性非IgA系膜增生性肾炎(MsPGN)、原发性局灶节段性肾小球硬化症(FSGS)和狼疮肾炎(LN)患者肾组织的表达及其意义。方法采用原位杂交技术(ISH)检测34例上述3种肾脏病患者及4例对照组肾组织中IL-13mRNA表达量的变化,分析IL-13基因表达量与肾功能变化的关系。结果IL-13mRNA在FSGS、MsPGN和LN患者肾组织内表达均增高,而对照者肾组织内无明显IL-13mRNA表达。LN患者肾小管间质区IL-13mBNA表达水平与狼疮活动相关,MsPGN及FSGS患者肾小管间质区IL-13mRNA表达量与肾功能变化及24h尿蛋白量相关。结论IL-13可能直接参与人类肾小球疾病的免疫反应过程。

Abstract:Objective To study the clinical significance of interleukin- 13 (IL-13) gene expression in renal tissue of human kidney diseases.Methods 4 controls and 34 patients with primary focal segmental glomerular sclerosis(FSGS),non-IgA primary mesangioproliferative glomerulonephritis(MsPGN) and lupus nephritis(LN) were included in this study.IL-13mRNA expression in renal tissue was detected by in situ hybridization(ISH) technique.Results Up-regulating expressions of IL-13mRNA in the renal tissues of FSGS,MsPGN and LN were found,whereas little expression of IL-13mRNA in the renal tissue of the controls was observed.The elevated expression of IL-13mRNA in tubulointerstitium was closely related to lupus activity in LN.The levels of IL-13mRNA in tubulointerstitium correlated to the renal function and urinary albumin excretion in FSGS and MsPGN.Conclusion IL-13 may be involved in the immune reaction of renal tissue in human kidney diseases.
