Effects of ligustrazine on the peritoneal membrane function and the mesothelial cell morphology of chronic peritoneal dialysis rats

Abstract:Objective To investigate the influence of different concentrations of ligustrazine on the cell morphology and the peritoneal membrane function and their relationship in chronic peritoneal dialysis rats with infusion of high glucose dialysate. Methods 40 SD rats were divided randomly into 4 groups. Except control group, other groups daily received infusion of 20 ml 4.25% glucose dialysate containing different concentrations of ligustrazine [0 (HG), 40 mg/L (HGL), 160 mg/L (HGH)] respectively for 8 weeks. The peritoneal membrane function was investigated by peritoneal transport test , and the rat peritoneal mesothelial cells(PMCs)morphology was analyzed from the cell imprints. Results Compared to the HG and HGH groups, the intraperitoneal volume and net ultrafiltration in HGL group were significantly higher ( P< 0.01), D/Purea significantly lower, and D/D0 and Curea after 4 hours of dialysis significantly higher. The peritoneal fluid absorption rate as well as direct lymphatic absorption rate had no significant difference between all the groups. The density of cell population from analysis of cell imprints in HGL group was significantly higher than that of HGH and HG groups, but the mean surface area was significantly lower. Long-term use of low dose ligustrazine in the high glucose dialysate could significantly increase the ultrafiltration volume, which was significantly correlated to the decreasing of cell surface area (r=-0.944, P<0.05) of PMCs. Conclusion Low-dose ligustrazine added to high glucose dialysate can significantly increase the ultrafiltration volume, which might related to the decrease of the hypertrophy of PMCs.
