NF-κB抑制剂对THP-1粘附及MMP-9表达影响的实验研究

摘要前单核细胞THP-1分化是一个复杂的过程,这一过程包含了复杂的细胞内信号传导及许多因子的活化及大量蛋白的表达。基质金属蛋白酶-9(Motrix Metalloproteinase-9, MMP-9)主要降解基底膜中的Ⅳ和Ⅴ型胶原,在血管增生及肿瘤浸润转移过程中起重要的作用。为探讨核转录因子κB(nuclear factor kappa B, NF-κB)在THP-1粘附及MMP-9表达中的作用,我们应用NF-κB抑制剂PDTC(Pyrollidine Dithiocarbamate二硫碳吡咯烷,应用共聚焦显微镜、流式细胞术观测了其对THP-1粘附及MMP-9表达的影响。结果表现:PDTC可明显的抑制THP-1的粘附及MMP-9的表达。表明NF-κB在细胞粘附及MMP-9的表达中起重要的作用。

AbstractThe differentiation of pro-monocyte THP-1 is a complicated process which includes the activation of many transcription factors such as NF-κB and the cxpression of many proteins such as adhesion molecules. In the article, we use the inhibitor of NF-κB, pyrollidine dithiocarbamate (PDTC) to explore the effect of PDTC on the THP-1 adhesion and MMP-9 expression measured by flow cytometry. The results suggest that the inhibitor of NF-κB inhibits the adhesion of THP-1 and partly inhibits MMP-9 expression.
