Laminin是一个巨大的糖蛋白家族，是构成细胞外基质(extracellular matrix，ECM)的主要成分，通过直接和间接地与其它细胞外基质蛋白质如四型胶原蛋白，硫酸肝素，nidogen/entactin，proteogly-can[1，2]，等的相互作用形成网状结构，构成基底膜[3]。研究显示Laminin通过与细胞表面受体相互作用影响细胞的粘附、转移、生长和分化[4]，因此，Laminin在细胞的发育，组织的形成及基底膜的机械支撑等方面起着重要的作用。Laminin对神经细胞的作用尚不完全清楚，中枢神经系统与其它系统组织不同，被认为缺乏细胞外基质。在高等脊椎动物，由于分化成熟的神经元不能再进入细胞分裂周期，而无再生能力，而且，在中枢神经系统，受损的轴突很难再生来恢复与靶细胞的连接能力。然而，周围神经系统有细胞外基质，并且其轴索也有一定程度的再生能力。细胞外基质蛋白质对神经再生的作用已经在体外神经细胞培养中得到证实。在本研究中，我们克隆了线虫C、elegans的Laminin α链基因并测定了其基因组及cDNA序列，推算出了Laminin α链氨基酸序列，同时我们分离了Laminin α基因突变的变异体并决定了变异体的变异部位。利用绿荧光蛋白(GFP)标记物，观察了野生型和变异体的神经细胞异常发育的异常形态改变，这些研究证明了Laminin α链基因突变引起了神经系统异常发育，对细胞外基质成分和神经组织的相互作用，进而对人类神经系统疾病研究提供了基础。……
