The levels of plasma IL-1β,IL-6,corticosterone of Balb/c mice with lateralization

Abstract:Objective: To study the relationship between cytokines,HPA axis and lateralization.Methods:By using paw preference test,right-pawed ,left-pawed,ambidextrous mice model were established. ELISA kits and radioimmunoassay were used for detection of plasma levels of cytokines and corticosterone.Results:In untreated normal Balb/c mice ,left-pawed mice had a higher IL-1β level than right-pawed mice (P<0.05),while IL-1β of ambidextrous was the highest among them; There was no difference observed between the three groups in plasma levels of IL-6; As for plasma levels of corticosterone in untreated mice, left-pawed and right-pawed mice were both lower than ambidextrous mice.In LPS treated mice, plasma levels of IL-1β were higher in left-pawed mice than in right-pawed and ambidextrous mice, and right-pawed mice was lower than ambidextrous; The plasma levels of IL-6 were lower in left-pawed and right-pawed mice than in ambidextrous(P<0.01). The plasma levels of corticosterone were almost the same among the three groups after the injection of LPS. Conclusion: Lateralization could influence cytokine levels and HPA axis in mice.

机体在发育过程中形成了脑不对称性(Brain lateralization)。脑不对称性是一个普遍的生物学现象,对机体许多功能有明显影响。因手功能的不对称性是脑不对称性最明显的表现,研究上常按手功能不对称性的不同将人群区别为惯用左手者(左利,Left -hander,LH),惯用右手者(右利,Right-hander,RH),无明显惯用手者(双利,Ambidextrous-hander,AH)。自从80年代Geschwind报道了左利人群有较高的免疫紊乱发生率后,研究者运用伸爪取食法区分出左利和右利动物, 深入研究脑不对称性对免疫功能影响及可能的机制。正常状况下左利和右利动物T细胞、Mφ细胞、NK细胞活性不同,用LPS腹腔注射动物模拟细菌感染和炎症性疾病的情况下,左利和右利动物血浆ACTH和皮质酮(Corticosterone,CS)上升幅度亦与脑不对称性有关[1]。神经免疫内分泌网络在免疫紊乱发病机制中起的作用,越来越受重视,IL-1、IL-6和CS是其中的重要因子。为探讨左利者免疫紊乱发生率高的原因,有必要研究脑不对称性对神经免疫内分泌网络的影响。为此,本文研究了脑不对称性Balb/c小鼠正常状况及受LPS 刺激后血浆IL-1β、IL-6和CS的水平变化。
1材料与方法
1.1实验动物Balb/c健康小鼠,雌性,4周龄,由中山医科大学实验动物中心供应。在本实验室喂养,5只/笼,自由饮水取食,每天保持12 h明暗环境,行为测试及最后的实验均在上午进行。
1.2左利和右利动物模型的建立动物适应环境1 w后经伸爪取食法区分为左利、右利和双利3组。方法如下:小鼠禁食12~18 h后,置于一特制盒内,盒子体积为4.5 cm×4.5 cm×6.0 cm,前壁中央连有一小管可供放入小饲料块,以小饲料块逗引小鼠伸爪取食,观察其50次伸爪取食中伸右爪次数,记为右利分(Right paw entry,RPE)。2 w内共测试4次,第1次作为训练,不记录,将余下3次右利分平均,0~20分为左利鼠(Left-pawed mice),21~29分为双利鼠(Ambidextrous mice),30~50分为右利鼠(Right-pawed mice)[2]。区分后1 w,实验组每只小鼠以LPS(Escherichia coli serotype 026:B6, Sigma) 5 μg/ 0.5 ml无菌生理盐水,正常对照组用0.5 ml无菌生理盐水(NS),腹腔注射。注射后2 h和4 h分别快速断头杀鼠。用置于冰上并肝素化的试管收集小鼠血液,3 000 r/min离心15 min后取血浆,分装后置于-30℃保存待测。
1.3血浆IL-1β和IL-6检测用ELISA法检测,使用由深圳晶美公司提供的ENDONGEN公司生产的小鼠IL-1β、IL-6 ELISA试剂盒。按说明书使用,A值用SPSS软件绘出标准曲线,计算各样本的值。
1.4血浆CS检测肝素抗凝血浆10 μl加无水乙醇240 μl 于Eppendorf管,振荡混旋10 s,10 000 r/min 离心10 min,取上清50 μl入一小玻璃试管,每个样品设2管,40℃,15 min真空离心干燥,即为制备的样品管。同时设已知CS含量的质控管,用3H-CS、CS结合蛋白进行放免测定。
1.5统计方法用SPSS和EXCEL软件对各组血浆IL-1β、IL-6、CS水平进行单因素方差分析(ANOVA)。
2结果
2.1Balb/c 小鼠脑不对称性分布模式(图1) 左利小鼠28只(RPE在0~20之间),右利小鼠30只(RPE在30~50之间),双利小鼠18只(RPE在21~29之间)。左利小鼠的RPE主要在15~20间,右利小鼠的RPE在35~40间。

2.2正常对照组血浆IL-1β和IL-6水平未注射LPS的正常对照组血浆IL-1β水平测定结果表明(图2),左利组显著高于右利组(P<0.05),双利组显著高于左利、右利组(P<0.05)。其血浆IL-1β水平依次为双利组>左利组>右利组。血浆IL-6 如图3所示,正常对照各组之间血浆IL-6水平依次为右利>双利>左利,但均无统计学意义。

2.3LPS注射后血浆IL-1β和IL-6水平变化血浆IL-1β 如图2所示,LPS刺激后2 h,左利组显著高于右利组(P<0.05),双利组亦显著高于右利组(P<0.05),左利与双利组间无明显差异;刺激后4 h,左利组和双利组仍然显著高于右利组(P<0.05)。左利组LPS刺激4 h后较2 h有所上升,而右利组和双利组IL-1β均较2 h组下降。LPS刺激后,左利组上升幅度最大,约为5倍(2 h)和7倍(4 h),右利组次之,约2.4倍(2 h)和2.0倍(4 h),双利组约1.5倍(2 h)和1.8倍(2 h)。由此可认为左利组对LPS刺激引起的IL-1β上升最敏感。血浆IL-6水平如图3所示,LPS刺激后2 h,左利组上升幅度约为6倍,右利组上升幅度约为7倍,双利组升高了约18倍,显著高于左利组和右利组(P<0.01), 双利组对LPS刺激引起的IL-6上升最敏感。LPS刺激后4 h,左利、右利和双利3组间IL-6无明显差异。与LPS刺激后2 h相比,左利、右利和双利3组IL-6在刺激后4 h均显著下降(P<0.05)。

2.4LPS注射后血浆CS水平变化见图4,未注射LPS正常对照双利组血浆CS水平显著高于左利和右利组(P<0.05),左利组血浆CS水平亦高于右利组,但两组间差别无统计学意义。LPS刺激2 h后,左利、右利和双利3组血浆CS水平均明显升高(P<0.05),但3组间血浆CS水平无明显差别。
