Effect of recombinant interferon-γ(rIFN-γ)on IL-8 and IL-10 released by peripheral blood mononuclear cells(PBMC)in asthmatic patients

Abstract:Objective To study the effect of (rIFN-γ)on IL-8 and IL-10 release in asthmatic patients PBMC and its relationship with serum level of ECP.Methods Samples of PBMC obtained from 8 cases of asthmatic patients and five cases of normal persons were cultured in RPMI 1640 containing 10% bovine serum.After 66 hours,the supernatants of cultured PBMC of asthmatic patients were divided into three groups:group A (rIFN-γ),group B (Dexamethasone)and group C (control group).Concentration of IL-8 and IL-10 in the supernatant in the three groups was determined after 72 hours.Serum ECP levels were also determined.Results The serum ECP level was positively and signifocantly correlated with content of IL-8 in the supernatant (r=0.701)and negatively correlated With that of IL-10(r=0.638).There was significant difference between the serum ECP level and the IL-8 and IL-10 concentration in supernatants of cultured PBMC (P<0.05).After rIFN-γ intervention ,the IL-8 level decreased significantly (P<0.05)and the IL-10 increased singificantly(P<0.05)than that of control group.There was no significant difference after dexamethasone intervention.Conclusion rIFN-γ may decrease the level of IL-8 which had anti-inflammation effects.IL-10 may inhibit the proliferation and differentiation of inflammatory cells caused by antigen stimulation and may inhibit the synthesis of cytokines and inflammatory mediators and also inhibit B cells to secret IgE.rIFN-γ can promote the synthesis and secretion of IL-10 which can increase its anti-inflammatory function.
