Expression of vascular endothelial growth factor and its relationship with release of vascular active substances in placentae of patients with pregnancy induced hypertension

Abstract:ObjectiveTo investigate the expression of placental vascular endothelial growth factor (VEGF) and its relationship with placental release of prostacyclin I2 (PGI2) and thromboxane A2(TXA2) in patients with pregnancy induced hypertension (PIH).Methods Twenty-five patients with PIH and twenty-five normal late pregnant (NLP) women in the third trimester were observed in this study. The expression of VEGF protein in placenta, the plasma and placental homogenate levels of PGI2 and TXA2 were determined by Western blot and radioimmunoassay respectively. Results Compared with NLP, the expression of placental VEGF protein was significantly decreased (P<0.01). Plasma and placental homogenate levels of PGI2, PGI2/TXA2 in patients with PIH were significantly decreased (P<0.01) while TXA2 level was increased (P<0.01). There was a significant positive correlation between placental VEGF and PGI2 and PGI2/TXA2 (P<0.01). A significant negative correlation was found between placental VEGF and TXA2 (P<0.01), systolic (P<0.01)and diastole blood pressure(P<0.05). Conclusion It is suggested that the reduction of placental VEGF may down-regulate local PGI2/TXA2, resulting in the imbalance of maternal PGI2/TXA2 and dysfunction of endothelial cell.
